Solutions:

1. Phytohemagglutin (PHA) enriched RPMI-1640 media

2. Colcemid solution, 10ug per ml, in Hanks balance salt solution (HBSS)

3. Hypertonic KCl, .075M

4. Fixative, 3 parts anhydrous methanol to 1 part glacial acetic acid

5. Giemsa working solution 1% at pH 6.8

6. Giemsa stock solution

7. Phosphate buffer 0.025M (3.4 g KH2PO4 per liter adjusted to pH 6.8 with KOH)

8. Trypsin (1:250), 1% solution in water

9. Methanol

10. Acridine orange
